Ce film est la première partie d'une compilation en deux parties de l'anime Mawaru Penguindrum, comprenant des ajouts inédits. Après l'effondrement d’un aquarium, la sœur gravement malade de Shoma et Kanba, Himari, est ranimée grâce au chapeau de pinguin qu’elle désirait. Pour la garder en vie, les frères doivent retrouver un objet mystérieux appelé le Penguindrum. Dans leur quête, ils doivent résoudre des énigmes, s’allier de façon précaire et affronter des questions sur le temps et la destinée.
